Your tasks: Understanding the regulation of self-renewal, differentiation and metastasis formation of tumor-initiating cells (TIC) is pivotally important to eradicate malignant stem cell-like cells in human cancer and to develop safe and efficient therapeutic strategies.

We have demonstrated before that tumor-initiating cells within individual colorectal cancers functionally differ in their potential to self-renew and that distinct classes of TIC drive long-term tumor progression and metastasis formation.

Recent and current approaches in our lab employ large scale molecular targeting screens including vector driven insertional mutagenesis, pooled shRNA libraries and small molecule libraries as well as selective ribosome profiling in genetically defined TIC-enriched spheroid cultures. Using these approaches, candidate genes potentially regulating self-renewal, tumor-initiation or metastasis formation and novel substances with TIC inhibitory potential have been identified.

Within future projects, the interplay between genetic and functional tumor heterogeneity in human colorectal cancer will be investigated and selected candidate structures will be chosen for the development and validation of molecular strategies targeting tumor-initiating cell activity.

The National Center for Tumor Diseases (NCT) Heidelberg offers a highly interactive and collaborative scientific environment with a strong focus on translational cancer research.

Your profile: Applicants should hold a PhD degree in biology/life sciences and an excellent background in cellular and molecular biology. The applicants should be highly self-motivated and be able to pursue research projects independently as well as in collaborations.

Successful candidates will perform molecular and functional analysis of human malignant and non-malignant spheroid and organoid cultures. The major focus will be investigating genetic and functional heterogeneity and the molecular targeting of TIC in vivo.
