Job description

Het is aangetoond dat lactoferrine (LF) een rol speelt in ijzer homeostatis (Frazer et al., 2011), celgroei en –differentiatie (Bi et al., 2010), afweer tegen microbiële infecties (Atef Yekta et al., 2010), protectie tegen virale infecties (Wakabayashi et al., 2014) en anti-inflammatoire werkt (Actor et al., 2009). Wij toonden inhibitie aan van groei van E. coli O157:H7 stammen en van hun aanhechting aan intestinale epitheliale cellen (Atef Yekta et al., 2010). Naast deze effecten op virulentiefactoren, demonstreerden we ook in schapen en kalveren dat LF het immuunsysteem van de gastheer kan beïnvloeden. In herkauwers, zagen we verhoogde expressie van genen die het immuunsysteem activeren en verminderde expressie van genen die het immuunsysteem onderdrukken (Kieckens et al., 2016). Daarnaast traden er verhoogde antistoffenresponsen op onder invloed van LF en werd meer IgA gedetecteerd ter hoogte van de darmmucosa. In dit onderzoek zullen we nagaan of vergelijkbare effecten kunnen optreden bij biggen door varkens cellen en cellijnen met lactoferrine te incuberen en vervolgens effecten op het immuunsysteem na te gaan. Cytokines, genexpressie en reactie op een infectie met enterotoxigene E. coli zijn daarbij parameters die zullen geanalyseerd worden.
Je zal onderzoek doen in een uitdagende omgeving waar een stimulerende team spirit je zal helpen belangrijke onderzoeksvragen op te lossen. Valorisatie is een belangrijke doelstelling van dit onderzoek en het vertalen van bevindingen in dit onderzoek naar de praktijk is één van de belangrijke doelstellingen. Je zal de mogelijkheid hebben om in verschillende laboratoria te werken samen met toponderzoekers. Dit is veruiterlijkt in de nummer 1 positie die het diergeneeskundig onderzoek van onze faculteit heeft in de Shangai global ranking Veterinary Sciences.
It has been demonstrated that lactoferrin (LF) plays an important role in iron homeostasis (Frazer et aL, 2011), cell growth and cell differentiation (Bi et al., 2010), resistance against microbial infections (Atef Yekta et al., 2010), protection against viral infections (Wakabayashi et al., 2014) and has an anti-inflammatory effect (Actor et al., 2009). We showed inhibition of growth of E. coli O157:H7 strains as well as inhibitiono of their adhesion to epithelial cells (Atef Yekta et al., 2010). Besides, we demonstrated in sheep and cattle that LT modulates the host immune system. This was evidenced by increased expression of genes involved in immune stimulation and a decreased expression of genes involved in immunosuppression (Kieckens et al., 2016). In addition, increased antibody responses were observed following treatment with LF. The aim of the present research proposal is to examine if similar effects can be demonstrated in pigs. Hereto, in vitro studies will be performed using porcine primary cells and cell lines to demonstrate effects of LF on interaction of these cells with F4 enterotoxigenic E. coli and F18 verotoxigenic E. coli and analyze gene expression and cytokine responses.

  • You hold a master degree in Veterinary Medicine or in Bioscience Engineering.
